Using population isolates in genetic association studies
The use of genetically isolated populations can empower next-generation association studies. In this review, we discuss the advantages of this approach and review study design and analytical considerations of genetic association studies focusing on isolates. We cite successful examples of using population isolates in association studies and outline potential ways forward.
متن کاملPopulation Structure in Genetic Association Studies
Standard genetic association tests using case-control data are based on certain assumptions about the population from which study subjects were sampled. Two types of departure from these assumptions have been studied: population stratification and cryptic relatedness. Both types of departure have been called population structure. Each can lead to erroneous inferences due to differences between ...
متن کاملPopulation stratification and genetic association studies in South Asia
Population stratification and its influence on genetic association studies is a controversial topic. Although it has been suggested that stratification is unlikely to bias the results of association studies conducted in developed countries, convincing contrary empirical evidence has been published. However, it is in populations where historical ethnic, religious and language barriers exist that...
متن کاملPopulation Structure and Cryptic Relatedness in Genetic Association Studies
We review the problem of confounding in genetic association studies, which arises principally because of population structure and cryptic relatedness. Many treatments of the problem consider only a simple “island” model of population structure. We take a broader approach, which views population structure and cryptic relatedness as different aspects of a single confounder: the unobserved pedigre...
متن کاملCorrecting population stratification in genetic association studies using a phylogenetic approach
MOTIVATION The rapid development of genotyping technology and extensive cataloguing of single nucleotide polymorphisms (SNPs) across the human genome have made genetic association studies the mainstream for gene mapping of complex human diseases. For many diseases, the most practical approach is the population-based design with unrelated individuals. Although having the advantages of easier sam...
